High Performance Liquid Chromatography (HPLC) Method for Analysis of Biotin (Vitamin B7)

Biotin (Vitamin B7) is a water-soluble vitamin that helps your body breakdown key nutrients in order for it to absorb and utilize them. A lack of biotin can lead to hair loss, rash, and brittle nails. A healthy diet including eggs, milk, and bananas can supply your body with a healthy dose of biotin.
Using a Newcrom BH mixed-mode column and a mobile phase consisting of water and acetonitrile with an ammonium formate (AmFm) buffer, Vitamin B7 can be retained and measured. Vitamin B7 can be detected using charged aerosol detection (CAD) with high resolution.

Application Column

Newcrom BH

The Newcrom columns are a family of reverse-phase-based columns. Newcrom A, AH, B, and BH are all mixed-mode columns with either positive or negative ion-pairing groups attached to either short (25 Å) or long (100 Å) ligand chains. Newcrom R1 is a special reverse-phase column with low silanol activity.

Separation of Biotin on Newcrom R1 HPLC column

Separation of Biotin on Newcrom C18 HPLC column

Biotin, also known as vitamin B7, is a micronutrient produced by the body that helps break down carbohydrates, fatty acids. It also helps form sugar from non-carbohydrate substances.
Biotin can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible, applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particle columns are available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation of impurities in preparative separation. It also suitable for pharmacokinetics.
