HPLC Method for gamma-Aminobutyric Acid (GABA), Baclofen, Pregabalin, Vigabatrin, Phenibut on Primesep 100 by SIELC Technologies

High Performance Liquid Chromatography (HPLC) Method for Analysis of Gamma-aminobutyric acid (GABA), Vigabatrin, Phenibut, Pregabalin, Baclofen
Gamma-Aminobutyric Acid (GABA) is a neurotransmitter with the chemical formula C4H9NO2. It is crucial in the nervous system for reducing anxiety and promoting sleep.
Vigabatrin is a medication with a chemical formula C6H11NO2. In Canada, it is approved for use as an adjunctive treatment for epilepsy, seizures, , and spasms in West syndrome.
Phenibut is a central nervous system (CNS) depressant with the chemical formula C10H13NO2. It is prescribed primarily in Russia and greater Eastern Europe to treat anxiety, insomnia, and other conditions.
Pregabalin is a medication with the chemical formula C8H17NO2. It is an anticonvulsant, analgesic, and anxiolytic amino acid that is used to treat epilepsy, restless leg syndrome, anxiety, neuropathic pain, and fibromyalgia. In the US it is used for adjunctive therapy for adults, while in the European Union, United Kingdom, and Russia, it is approved for treatment of generalized anxiety.
Baclofen is a central nervous system (CNS) depressant with the chemical formula C10H12ClNO2. It is used to treat spastic movement disorders. Off-balel, it is used as treatment for alcohol use disorder and opioid withdrawals.
All gabapentinoids can be retained and separated on the Primesep 100 mixed-mode column using an isocratic analytical method with a simple mobile phase of water, acetonitrile (MeCN, ACN), and Ammonium Formate (AmFm) buffer. Detection can be achieved with mass spectrometry (MS), evaporative light scattering detection (ELSD) and Charged aerosol detection (CAD).

Separation of Baclofen on Newcrom R1 HPLC column
February 16, 2018
Baclofen can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.
Application Column
Newcrom R1
The Newcrom columns are a family of reverse-phase-based columns. Newcrom A, AH, B, and BH are all mixed-mode columns with either positive or negative ion-pairing groups attached to either short (25 Å) or long (100 Å) ligand chains. Newcrom R1 is a special reverse-phase column with low silanol activity.
