High Performance Liquid Chromatography (HPLC) Method for Analysis of Pseudoephedrine (PSE), Fexofenadine hydrochloride.
Pseudoephedrine is one of the most popular decongestant drugs on the market, in part due to its efficacy and in part due to its status as an amphetamine. It has the chemical formula C10H15NO. It has been reported to have been misused as a psychostimulant, since it is accessible over-the-counter. It was a banned substance by the International Olympic Committee (IOC) until 2004, but was banned again in 2010.
Fexofenadine is a common antihistamine with the chemical formula C32H39NO4. It is used to treat hives, itching, and hay fever. Pseudoephedrine and Fexofenadine can be separated, retained, and analyzed on a Primesep 100 mixed-mode column using an isocratic analytical method with a simple mobile phase of water, Acetonitrile (MeCN), and a sulfuric acid (H2SO4) buffer. This analysis method can be UV detected at 200 nm with high resolution and peak symmetry.

Separation of Fexofenadine hydrochloride on Newcrom R1 HPLC column

Fexofenadine hydrochloride can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.
