HPLC Method for Analysis of Denatonium Benzoate on Primesep SB Column
June 30, 2022
Denatonium Benzoate is the most bitter compound ever discovered. It has several popular applications, including as a key ingredient in denaturing ethanol (for tax and sales purposes), as a placebo to mimic the bitter taste of certain pills, and as a biochemical warning to prevent the consumption of various types of harmful organic substances (including methanol, acetone, and paint, among others). Denatonium Benzoate can be separated and retained on a mixed-mode Primesep SB column with embedded strong basic ion-pairing groups, using a gradient analytical method with a simple mobile phase of water, Acetonitrile (MeCN), and an Ammonium Formate (AmFm) ionic modifier. This analysis method can be UV detected at 260 nm with high resolution and peak symmetry and is compatible with Mass Spectrometry (MS), ELSD, and CAD.

High Performance Liquid Chromatography (HPLC) Method for Analysis of Denatonium benzoate, Benzoic Acid
Denatonium benzoate, also known as Denatonium, is the bitterest chemical that is safe to be ingested. It is often used as a bittering agent to antifreeze, detergents, and cosmetics to prevent consumption. Even low concentrations of it can cause am incredibly bitter taste. In France and Italy it is mandatory to add Denatonium to antifreeze. It has the chemical formula C₂₈H₃₄N₂O₃.
Benzoic Acid is an organic compound with the chemical formula C7H6O2. It is considered to be the simplest aromatic carboxylic acid. In nature, it can be found in many plants, especially berries. It is used for food preservation of food, as it can inhibit growth of bacteria and mold. While it is safe in food, it can cause irritation to skin, eye, and respiratory systems.
Denatonium benzoate, Benzoic Acid can be retained and analyzed using the Newcrom A stationary phase column. The analysis utilizes an isocratic method with a simple mobile phase consisting of water and acetonitrile (MeCN) with a sulfuric acid buffer. Detection is performed using UV.

Separation of Denatonium benzoate on Newcrom R1 HPLC column
May 16, 2018
Denatonium benzoate can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.
Application Column
Newcrom R1
The Newcrom columns are a family of reverse-phase-based columns. Newcrom A, AH, B, and BH are all mixed-mode columns with either positive or negative ion-pairing groups attached to either short (25 Å) or long (100 Å) ligand chains. Newcrom R1 is a special reverse-phase column with low silanol activity.
