High Performance Liquid Chromatography (HPLC) Method for Analysis of Octanoic acid (Caprylic acid).
Octanoic acid (Caprylic acid) is the common name for the eight-carbon straight-chain fatty acid. It can be found in numerous foods such as coconuts, cherries, plums, pineapple sages, black raspberries, and shallots. Octanoic acid is found to be associated with medium-chain acyl-CoA dehydrogenase deficiency, a genetic disorder that affects the metabolism of converting certain fatty acids to energy.
Octanoic acid can be retained on a mixed-mode Primesep 100 column by using an isocratic method consisting of a 50/50% Acetonitrile (ACN) and water mobile phase with a Phosphoric acid (H3PO4) buffer. UV detection at 210 nm.

Separation of Octanoic acid on Newcrom R1 HPLC column

Octanoic acid can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.
