High Performance Liquid Chromatography (HPLC) Method for Analysis of Ketorolac.
Ketorolac is a nonsteroidal anti-inflammatory drug (NSAID) with the chemical formula C15H13NO3. It is used primarily used to treat moderate to severe pain. Due to a high risk of negative effects, including mortal gastrointestinal bleeding, it is only taken for short intervals of time. Despite the adverse effects, it is a widely available and often prescribed in the United States.
This application describes a normal-phase chiral HPLC method for the separation of Ketorolac enantiomers using SIELC’s Cyralis CM chiral column. Cyralis CM is a cellulose-based chiral stationary phase (CSP) designed for enantiomeric separations, optical purity testing, chiral impurity analysis, and stereoisomer characterization in pharmaceutical, chemical, agrochemical, and fine chemical applications. For best results, the sample should be prepared fresh for analysis.
Ketorolac can be retained and analyzed using the Cyralis CM stationary phase column. The analysis utilizes an isocratic method with a mobile phase consisting of Hexane, Ethanol, and TFA. Detection is performed using UV.

Separation of Ketorolac on Newcrom R1 HPLC column

Ketorolac can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.
