HPLC Chiral Method for Analysis of Bisoprolol Enantiomers on Cyralis CM Column by SIELC Technologies

High Performance Liquid Chromatography (HPLC) Method for Analysis of Bisoprolol.

Bisoprolol is a beta-blocker with the chemical formula C18H31NO4. It is used primarily used to treat high blood pressure, heart failure, and chest pain. It works through targeting receptors, specifically epinephrine, in the heart, which slows heart rate and lowers blood pressure.

This application describes a normal-phase chiral HPLC method for the separation of Bisoprolol enantiomers using SIELC’s Cyralis CM chiral column. Cyralis CM is a cellulose-based chiral stationary phase (CSP) designed for enantiomeric separations, optical purity testing, chiral impurity analysis, and stereoisomer characterization in pharmaceutical, chemical, agrochemical, and fine chemical applications. For best results, the sample should be prepared fresh for analysis.

Bisoprolol can be retained and analyzed using the Cyralis CM stationary phase column. The analysis utilizes an isocratic method with a mobile phase consisting of sodium Hexane, Ethanol, and DEA. Detection is performed using UV.

Separation of Bisoprolol on Newcrom R1 HPLC column

Separation of Bisoprolol on Newcrom C18 HPLC column

Bisoprolol can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.

Application Column

Newcrom R1

The Newcrom columns are a family of reverse-phase-based columns. Newcrom A, AH, B, and BH are all mixed-mode columns with either positive or negative ion-pairing groups attached to either short (25 Å) or long (100 Å) ligand chains. Newcrom R1 is a special reverse-phase column with low silanol activity.
