4-Aminophenol (also known as para-aminophenol or Rodinal) is a popular film developing compound and building block in organic chemistry reactions. Most notably, it is a key compound used in the production of acetaminophen (also known as paracetamol or Tylenol). 4-Aminophenol can be separated, retained, and analyzed on a Primesep 100 mixed-mode stationary phase column using an isocratic analytical method with a simple mobile phase of water, Acetonitrile (MeCN), and an sulfuric acid (H2SO4) or Perchloric acid (HClO4)buffer. This analysis method can be detected in the UV regime at 275 nm.

This analysis method can be detected in the UV regime at 275 nm, and due to the Ammonium Formate buffer, is also compatible with evaporative detectors such as Evaporative Light Scattering Detectors (ELSD), Charged Aerosol Detection (CAD), and Electrospray Ionization (ESI) for Mass Spectrometry (MS).

Separation of 4-Aminophenol hydrochloride on Newcrom R1 HPLC column

4-Aminophenol hydrochloride can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.
