Ergocalciferol (Vitamin D2) and Cholecalciferol (Vitamin D3) are fat soluble vitamins that help your body absorb calcium and phosphorus. Your body can produce both when exposed to the sun, but you can also intake these essential vitamins through a daily multivitamin or a Vitamin D pill.
Vitamins D2 and D3 can be detected in the high UV regime. Using a Primesep SB reverse-phase column and a mobile phase consisting of water and acetonitrile with an acetic acid buffer, Vitamins D2 and D3 can be retained and separated. This analysis method can be UV detected at 265 nm.

Separation of Ergocalciferol on Newcrom R1 HPLC column

Ergocalciferol can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.
