High Performance Liquid Chromatography (HPLC) Method for Analysis of Benztropine methylsulfonate.
Benztropine is a medical drug used in treatment of patients with Parkinson’s disease. It works as an acetylcholine blocker decreasing muscle stiffness. Benztropine can be retained in HPLC on Primesep SB mixed-mode column by hydrophobic and anion-exchange properties. The isocratic method uses a mobile phase of acetonitrile (ACN) and water with sulfuric acid buffer and UV detection at 220nm.

Separation of Benztropine methylsulfonate on Newcrom R1 HPLC column

Benztropine methylsulfonate can be analyzed by this reverse phase (RP) HPLC method with simple conditions. The mobile phase contains an acetonitrile (MeCN), water, and phosphoric acid. For Mass-Spec (MS) compatible applications the phosphoric acid needs to be replaced with formic acid. Smaller 3 µm particles columns available for fast UPLC applications. This liquid chromatography method is scalable and can be used for isolation impurities in preparative separation. It also suitable for pharmacokinetics.
