High Performance Liquid Chromatography (HPLC) Method for Analysis of CBD, CBN, delta-9 THC, delta-8 THC

Cannabidiol (CBD), cannabinol (CBN), and delta-9-Tetrahydrocannabinol (delta-9 THC) are three of the most active cannabinoids in cannabis. Delta-8-tetrahydrobannabinol also occurs naturally, but in such small amounts that it is often cheaper and easier to synthesize it in the laboratory from CBD. Both forms of THC are the strongest of the cannabinoids and are the main cause of the psychoactive effects of cannabis. CBD, while the 2nd most active component, does not cause the same effects by itself, but studies show it may have other neurologic and psychiatric benefits. There has been a lot less research conducted on CBN and its potential health benefits, but it is a product of the breakdown of THC and works on the same receptors (albeit with different effects) as CBD.
These cannabinoids can be detected in the low UV regime. Using a Cannsep C reverse-phase column and a mobile phase consisting of water and methanol (MeOH) with no buffer, CBD, CBN, delta-9 THC, and delta-8 THC can be separated, measured, and analyzed. This analysis method can be UV detected at 275 nm with high resolution.
