Primesep B2 shows superior efficiency, symmetry, and peak shape for a hydrophilic basic compound compared to two leading reversed-phase C18 columns. Primesep B2 contains an embedded basic functional group that shields the underlying silica by ionic repulsion with the analyte. On reversed-phase columns exposed active silanols provide unintended polar interactions with basic analytes that lead to tailing. The HPLC separation uses a simple mobile phase of water, acetonitrile (MeCN, ACN) and trifluoracetic acid (TFA) with UV detection at 250 nm.
